LPS - Literature review Example The LPS of this group of bacteria can be divided into 3 components, 1) lipid A, 2) a core oligosaccharide, which can be further subdivided into an inner and outer core structure, and a glycosyl polymer of repeating units, called O-polysaccharide. The hydrophobic lipid A domain of the molecule contributes to the structural stability of LPS. The core oligosaccharide, on the other hand, maintain the semi-permeable barrier of the bacterial cellââ¬Ës membrane by cross-linking with divalent cations or polyamines (Frirdich and Whitfield, 2005). Because of its biological effects, it has been used in experimental research as a prototype endotoxin as well as an adjuvant. Through the elucidation that LPSââ¬â¢ lipid A component is a nontoxic immunostimulatory molecule, it was established that the lipid A domain modulates LPSââ¬â¢ biological activity (Harsoliya et al., 2011). According to Frirdich and Whitfield (2005), this domain of LPS is the most conserved, followed by the inner core oligosaccharide moiety attached to lipid A. 2. Modifications Just like other membrane proteins, LPS undergo structural changes to accommodate environmental changes such as availability of nutrients and balance of ions. In particular, lipid A modifications were found to enable the bacteria to adapt to an environment with low divalent cations, abundant cationic peptides and low temperature (Frirdich and Whitfield, 2005). The TLR4 pathway TLR4 is a membrane protein expressed by hematopoietic-derived, immune cells such as macrophages, neutrophils, lymphocytes and dendritic cells. It is the link of LPS to the innate immune pathway, which allows a faster recruitment of immune cells to the site of infection. It is able to do so by stimulating the transfer of nuclear factor- (NF-) to the nucleus, thus initiating the expression of the genes coding for IL-6, IL-1 and TNF-? (Creely, 2007), whose importance will be discussed in detail later. Because many body surfaces and mucosa are susceptible to bacterial infection, many body parts may be exposed to LPS as well. As such, they are also expressed by non-hematopoietic cells such as epithelia and endothelia. In the respiratory tract, many studies have already recognized the role of TLR4 in inducing defensive mechanisms against pulmonary infection. Since the non-hematopoietic cells do not have immune functions, they release signals that allow the recruitment neutrophils to the site of infection. If uncontrolled, however, this response may lead to a decrease in lung function due to inflammation of the lungs, as well as exacerbation of allergic asthma. (Hollingsworth et al., 2005). However, it was determine by Hollingsworth et al. (2005) that, at least in the respiratory tract, immune response is different in magnitude when LPS is detected by hematopoietic cells than if it is by epithelial and endothelial cells of the airway. In their experiment, they used chimeric mice to limit expression of TLR4 in 1) hematopoietic cells onl y, or in 2) structural cells only. Number of neutrophils present, cytokine/chemokine production, and airway inflammation, through assessment of tracheal pressure. AARON BURR Although Aaron Burr, b. Newark, N.J., Feb. 6, 1756, fought in the American Revolution and became an important political figure, serving a term (1801-05) as vice-president of the United States, he is best remembered today for having killed Alexander Hamilton in a duel. The son of a president of the College of New Jersey (now Princeton University) and the grandson of another (Jonathan Edwards), Burr could trace his ancestry back to the earliest Puritans. He entered Princeton at the age of 13, graduated at 16, and went on to become a Revolutionary War hero, rising to the rank of lieutenant colonel at the age of 21. In July 1782 he married Theodosia Bartow Prevost, the widow of a former British officer. They moved to New York City, where Burr built a reputation as an excellent attorney and made important political connections. He was "the most rising young man in the state," a contemporary noted. Political Career In 1789 Burr was appointed attorney general of New York by Gov. G eorge Clinton. Two years later the state assembly, which was controlled by partisans of Clinton and Robert Livingston, elected Burr to the U.S. Senate. His career in the Senate was not particularly memorable. Hamilton hated him, Clinton soon learned to distrust him, and George Washington refused his request to be appointed minister to France. But in and out of Congress, Burr managed to maneuver so skillfully, and with so much personal charm, that he won the support of many Federalists as well as Democratic Republicans. In 1796 and 1800, Burr ran for vice-president with Thomas Jefferson on the Democratic-Republican ticket. Whatever doubts Virginia Republicans had about Burr--they had not voted for him in 1796--were put to rest when he carried New York City for his party in 1800. It was assumed that the outcome of the national election would follow that in New York, but under the confused electoral system then in use Jefferson and Burr received an equal number of electoral votes for t he presidency (73 each), throwing the election into the House of Representatives. There the Federalists refused to heed the advice of Hamilton and unsuccessfully tried, against the obvious wishes of the public, to elect Burr. Jefferson won the contest and Burr became vice-president. Jefferson doubted his loyalty and soon began to withhold patronage from Burr a... ...a busy and lucrative law practice. He continued to have a strong influence in the public councils, however. He supported a defiant posture toward France during the XYZ AFFAIR (1798), and as inspector general of the army (1798-1800) he took charge of organizing the nation's defenses. Bitterly disappointed in President John ADAMS's erratic leadership, Hamilton openly opposed Adams's reelection in 1800. When it appeared, however, that Aaron Burr might win the presidency over Jefferson, Hamilton unhesitatingly threw his support to Jefferson, whose policies he scorned, rather than to Burr, whom he regarded as a man without principles. This and other opposition by Hamilton so frustrated and angered Burr that he challenged Hamilton to a duel. The two men fought at Weehawken, N.J., on July 11, 1804. Hamilton apparently fired into the air, but Burr took direct aim. Hamilton fell mortally wounded and died the next day in New York. He was buried in Trinity churchyard, New York City. He left his wife and seven children heavily in debt, but friends soon paid off the debts. Hamilton was mourned by his countrymen as one who had devoted his life to the nation's growth in freedom and prosperity.

HOW IT BEGAN: A Crash in Roswell, New Mexico ON-THE-SCENE EYEWITNESSES DESCRIBE CRASH AND UNUSUAL WRECKAGE On the night of July 4, 1947, William Woody, who lived east of Roswell, was outside with his father at their ranch, when they saw a brilliant fiery object plunge to the ground (IUFOMRC). On the morning of July 8th, Roswell Sheriff George Wilcox, who along with W. W. ââ¬Å"Macâ⬠Brazell, found the crashed object on the J. B. Foster sheep ranch near Corona, 85 miles northwest of Roswell. Brazell, who lived on a nearby ranch, had notified the sheriff the day before about the crash (AJ). Wilcox called Major Jesse Marcel, Intelligence Officer at the nearby Roswell Army Air Force Base[1], informing him of his find. Marcel, joined by Sheriff Wilcox and Sheridan Cavitt, Counter-Intelligence Officer from the Roswell base, raced to investigate the crash site (Jaroff). Hamlet: to Be, or Not to Be: Movie Analysis Essay
Branaghââ¬â¢s vision of William Shakespeareââ¬â¢s famous ââ¬Å"To be, or not to be,â⬠soliloquy manifests Hamletââ¬â¢s displeasure with himself as he debates eternal sleep; the set up of this scene contributes significantly to the emotional impact and symbolism. The lack of music and sound in the beginning forces the audienceââ¬â¢s attention towards the soliloquy. The quietness in the scene also exhibits how Hamlet is wishing for a quiet death, suicide. Walking slowly towards the two-way mirror, while he professes his conflicted feelings, the camera follows steadily over his shoulder, only filming Hamletââ¬â¢s reflection. The soliloquy stands alone as a reflection, Hamlet is reflecting on his life and the options he has, ââ¬Å"To die, to sleep ââ¬â / No more ââ¬â and by a sleep to say we endâ⬠(3.1 68-69). The mirror reflection stands as a symbolic example of Hamletââ¬â¢s self reflection and his search to find the answer within himself. Hamlet criticizes himself while staring deeply at his own reflection in mirror, even though he is aware of Claudius malicious acts. Blaming himself for not taking action yet, ââ¬Å"Thus conscience does make cowardsâ⬠(3.1 91), and focusing on his faults he is making himself to be the villain and tormenting himself. The true reason for his misery is Claudius actions, and Claudius, the one behind it all- metaphorically and physically- stands behind the mirror as Hamlet criticizes himself. Branaghââ¬â¢s tone of voice and body language exert emotions in the scene making Hamletââ¬â¢s true feelings apparent. Half way through the soliloquy, suspenseful classical music begins to play, adding intensity to Hamlets dark description of the unnecessary burden of life and his ambiguous understanding to why a person would endure such suffering, ââ¬Å"For who would bear the whips and scorns of time, / Thââ¬â¢ oppressorââ¬â¢s wrong, the proud manââ¬â¢s contumely,â⬠(3.1 78-79). There are few actions Hamlet makes in the scene but the movements he does make are significant. Calmly stating, ââ¬Å"Or to take arms against a sea of troubles / And, by opposing, end them,â⬠(3.1 67-68) as he raises a clenched fist in the air, illustrating his anger toward Claudius and passion to seek revenge is portrayed through the small, but powerful hand gestures. It is indubitable that Branaghââ¬â¢s version of this scene captures both the emotions and themes of Hamlet and his tortured character.
